No. You measure the voltage that the starter sees while cranking. So you're measuring the running voltage of the starter motor, between the big cable as close to the starter motor as possible, and the body of the starter.

The tester leads...
red to cable from battery to starter (at starter)...
black to a ground on starter (starter body)...
